Path4Ward is a no-cost genetic testing program sponsored by X4 Pharmaceuticals in partnership with Invitae, intended to help diagnose U.S. patients suspected of having a congenital neutropenic disorder or a primary immunodeficiency (PID), including WHIM syndrome. The program includes genetic counseling for patients and/or clinicians, as well as family variant testing when applicable.

For patients suspected to have WHIM syndrome, genetic testing may help to:

  • Detect a CXCR4 variant known to cause WHIM syndrome
  • Inform a diagnosis to help guide treatment decisions
  • Provide information to support participation in clinical trials
  • Identify family members who may also be at risk
Individuals with a CXCR4 pathogenic variant have a 50% chance
of passing the disease to their offspring.

To qualify for Path4Ward, patients must meet all 3 of the following criteria:

  1. Suspicion of a congenital neutropenic disorder or a primary immunodeficiency.
  2. Any prior history or a current absolute neutrophil count (ANC) ≤1,000 cells/µL on multiple occasions that is not related to drugs or chemotherapy or secondary to viral infection.
  3. One or more of the following:
    • Personal or family history of recurrent and/or severe infection
    • Personal history of
      • Lymphopenia
      • Hypogammaglobulinemia
      • Refractory or recalcitrant warts
    • Family history of neutropenia

To order a test kit for your patient on Invitae.com, here are some helpful tips:

  • Secure 30–60 minutes in your schedule. You may save your progress, but completing the process in one sitting is recommended.
  • Have your NPI #, medical organization information, and details of the patient for whom you are ordering a test ready.
  • Enlist the help of your office administrator, or, if possible, complete the process together with your patient and/or their caregiver.
